PCR thermocycler calibration & preventive maintenance — common questions

Why calibrate a PCR thermocycler?
Amplification, melting, and enzyme kinetics are temperature-sensitive. A block that is off-target or non-uniform across wells causes failed or inconsistent reactions. Calibration verifies the block hits and holds the right temperatures evenly.
What does thermocycler calibration measure?
Block temperature accuracy at set points, well-to-well uniformity, and (where applicable) ramp behavior — measured with NIST-traceable probes and documented on a certificate.
Do you also perform preventive maintenance?
Yes. PM includes inspection, cleaning, function checks, and verification so the instrument runs reliably between calibrations — reducing surprise failures mid-run.
How often should thermocyclers be calibrated?
Annual calibration is common, with PM on a complementary schedule. Heavily used or clinical-grade instruments may warrant more frequent verification.
